COMPUTERS

The BRC laboratories with approximately 40 supported PCs are connected via a local area network. Computers on the network connect via 10-drop lines to the internet through the MBL server. The MBL Information Technology department currently houses the CIDIS, Pharmabase and www.biocurrents.org for BioCurrents as well as in excess of 100 Enterprise servers that are the infrastructure /foundation for the entire Enterprise of the MBL. The IT department has an expert staff that supports the educational program, administrative systems and research services. IT is the main support for the Informatics program (uBio) in taxonomy that has established global inroads for delivery of services. The network at MBL is robust with a gigabyte connection to the Internet and is a member of the Internet 2 community through our partners at the Woods Hole Oceanographic Institution.

IMAGING

TEM Zeiss 10CA 20-100KV TEM equipped with:
• reduced magnification (increased contrast) cartridge(+/- 40 degrees tilt and 360 degree rotation)
• port with video camera, monitor, recording interface
• standard specimen cartridge and goniometer specimen cartridge
• 3 1/4 X 4 inch sheet film camera
• AMT digital 1MP side mount camera
*The charge is $30 per hour with a $0.60 per negative charge.
*The stand-up darkroom for the microscope is equipped with a temperature maintained nitrogen burst film developer system and processing chemicals.

SEM JEOL JSM-840 1- 35KV SEM equipped with:
• goniometer specimen stage (X, Y,Z, and tilt translation; and continuous rotation)
• LaB6 electron source, liquid nitrogen pump baffle and anti contamination device
• PC based Orion digital image capture system
• Polaroid 4 X 5 camera and 60mm camera, video monitor and recording interface
• Oxford cryo stage, preparation chamber, and transfer device
• Tracor Northern elemental dispersive spectrophotometer (EDS)
• HKL electron backscatter diffraction (EBSD) detector
*The charge is $30 per hour for standard SEM and an additional $20 per hour for cryoSEM and EDS. Cost per hour does not include film but it does include processing chemicals in the attached darkroom.

LIGHT OPTICS LABORATORIES; including confocal and image analysis
Microscopic and photomicrographic equipment is available year round much of it through the courtesy of Carl Zeiss, Inc. The latest in optical microscopy equipment for such techniques as phase, dark-field, fluorescence, polarization, reflected light, and DIC are available.

Included are the Zeiss AxioImager Z1, Axioscope and Discovery stereo units with accessories for high-resolution color or black and white as well as low-light digital imaging and enhancement.Cameras available are the Zeiss MRm Axiocam, HRC Axiocam and HRm Axiocam. A camera lucida is also available for the compound Axioscope microscope. The AxioImager Z1 has Zeiss Vision and UIC MetaMorph imaging software. The Discovery dissecting microscope has the Zeiss Vision and UIC MetaMorph.

Zeiss ApoTome
The ApoTome is an innovative structured illumination slider installed on the AxioImager Z1 microscope. Provides improved image quality and enhanced contrast in conventional fluorescence microscopy. Deblurred 2D planes, even with thick specimens. Utilizes a high-end technique for artifact-free structured illumination. Flexible technique used for the entire spectrum of potential fluorochromes with the appropriate filter cubes. Standard light microscopy and image analysis is charged at $10.00 per hour.

CONFOCAL MICROSCOPY SYSTEMS

Zeiss LSM-510 NLO META - Inverted
The MBL has a Zeiss LSM-510 NLO with META detector laser scanning confocal system. The Zeiss 510 LSM has three independent simultaneous confocal channels for reflection and fluorescence and an extra channel for transmitted light. System is capable of standard and 2 photon excitation using the Coherent Chameleon laser. This highly automated system creates 8 or 12-bit 2048x2048 digital images using a Pentium PC based user interface. The Zeiss 510 LSM is configured on an inverted Axiovert 200 M microscope with a variety of objectives available and a complete incubator chamber system.

Zeiss LSM-510 NLO META - upright
The MBL has a LSM-510 NLO with META detector laser scanning confocal system configured on a fixed stage Axioskop 2FS microscope with a variety of objectives and a Sutter stage and manipulators. The Zeiss 510 LSM has three independent simultaneous confocal channels for reflection and fluorescence and an extra channel for transmitted light. System is capable of standard and 2 photon excitation using the Chameleon XR laser provided by Coherent. This highly automated system creates 8 or 12-bit 2048x2048 digital images using a Pentium PC based user interface.
